This is the only one of six XP machines doing so.

This is the only one of six > XP > machines doing so. You're right - it shouldn't be happening. Do you have any idea of what has changed in the last two weeks on your system? Whatever it is, it sounds like the app you are using to play your mp3 file has decided to "update" that modification date, which isn't right.

That was the first place I looked.

Copy the song to a CD and (attempt to) play from there. Something might fuss about being unable to write to the CD. That "something" may be the culprit.
