I have a new Dell optiplex 3060 with 250Gb SSD & 500GB HDD, the machine came with this. My question is as my total storage required is about 150GB should I use the SSD to store everything or use it for windows 10 OS and put everything else on the hard drive. If I put everything on the SSD would it slow the PC

Put everything on the 500 GB plus to an external HDD that way if something happens to the internal storage then you have a back up.

Yes make a backup to an external drive.

Use the SSD for your operating system and programs you use frequently, use the internal HDD for storage of photographs, videos and documents.

However you do not appear to have a lot of saved data and if there is room, there is no problem storing this on your SSD, it will not slow down your computer.

If you don't expect to increase your likely storage you can simply keep everything on the SSD then back it all up to the HDD. However I had a machine with almost the same spec as yours and found the SSD soon became full so I put in a M2 drive of 1tb and transferred to OS to that - now I have the old SSD as a files drive and the HDD as a backup for everything although my HDD is 2Tb not 500Gb. - Using the M2 and SSD for all the main work makes the machine pretty fast!

The solution proposed by Joddle is a possibility if you wish to spend money on an extra drive, however check the power requirement, specifically the current drain of the new drive before your do this as Dell often do not provide much of a margin for any extra power drain from their power supplies.
