I didn't change anything on the network. Windows cannot access the file gpt.ini for GPO CN={90EF7385-6653-411E-BFE9-46C1F6A7E623},CN=Policies,CN=System,DC=myDomain,DC=local,DC=com. Seems you have areplication problem because the DNS points only to itself. Or do you mean what happens for the clients? The only thing you have to check/change is that the clients DNS configuration also points to the second DC and that the second DC is also Global Catalog server.
