Confusion over Windows Defender.

Is Windows Defender suppose to be ON and Running in-conjunction WITH Microsoft Security Essentials ?  Or is Defender

SUPPOSE to off.  I read that Microsoft reported to a review of their anti-virus product, that Defender works

WITH Essentials !!  Can anyone clear this up for me ? Thanks.
